Basic Course Immunoblotting - Course E07
In this course, we show you how to detect individual proteins from a protein mixture by means of antibodies. The course comprises the following areas of main emphasis:
- Specific sample processing and optimization
- Polyacrylamide gel electrophoresis (SDS-PAGE)
- Blotting methods and their areas of application
- Optimization of first and second antibodies
- Detection methods and evaluation of analysis results
- Catalogue of measures for the avoidance of common errors and solution of problems
- Quality control methods (Coomassie staining and immunoprecipitation)
- Special applications: nuclear extracts and membrane stripping
- and much more
The biochemical backgrounds of the western blot technique, on the basis of which strategies for the optimization of the test parameters can subsequently be derived, are discussed in depth in a theoretical introduction. Particular emphasis is placed here on the comparative discussion of various methods and their areas of practical application, as well as measures to control the quality of the test stages and avoid typical errors and/or correct the problems that frequently occur when conducting western blot analyses. In a series of practical tests, you then acquire and develop the skills required for the successful conducting of analyses.
This course requires previous basic knowledge of molecular biology and the structure of proteins.
