Advanced Course Molecular Cloning - Course E06
This course communicates how to successfully clone DNA fragments into vectors while selecting a strategy aligned to your subsequent application.
The course focuses mainly on:
- Production and preparation of the DNA fragments for cloning
- Enzyme systems (restriction enzymes, isoschizomers, dephosphorylation)
- Cloning vectors and expression vectors
- Cloning and subcloning
- Reporter and selection systems
- Ligations and transformation techniques
- Latest methods of the genetic manipulation of organisms
- Troubleshooting
The biochemical backgrounds of cloning techniques, on the basis of which strategies for the optimization of the test parameters can subsequently be derived, are discussed in depth in a theoretical introduction. Particular emphasis is placed here on the comparative discussion of various methods and their areas of practical application, as well as measures to control the quality of the test stages and avoid typical errors and/or correct the problems that frequently occur when conducting cloning experiments. In a series of practical tests, you then acquire and develop the skills required for the successful conducting of experiments in your own projects.
This course requires basic knowledge of molecular biology.
