Basic Course Realtime PCR - Course E03
This basic course conveys the entire theoretical and practical knowledge required to successfully conduct realtime PCR analyses all the way through from the biological sample to the statistically substantiated result.
In addition to a comprehensive overview of the various realtime PCR techniques and their specific properties, all of the steps required for the various realtime PCR analyses are performed in the course of extensive practical experiments designed to demonstrate how the challenges of this method can be met successfully and how the efficiency of the method can be used professionally.
The course focuses mainly on:
- Introduction to the biochemical and technical fundamentals of quantitative realtime PCR
- Comparative overview of the various analysis techniques and available equipment
- Optimized sample processing and template acquisition
- Selection of test parameters and experiment planning
- Primer and probe design
- Absolute and relative quantification by means of realtime PCR
- Detection and reference dyes
- Sequence-specific and non-sequence-specific detection
- Probe libraries and commercially available assays
- Optimized preparation and execution of complex realtime PCR experiments
- Melting curve analysis
- Standard curves and endogenous controls
- Presentation and processing of experimental data
- Statistical models for evaluating experimental data
- Catalogue of measures for avoiding and solving frequently occurring problems
- Special realtime PCR applications in analysis and diagnostics
- Discussion forum
In addition to intensive theoretical introductions to the individual groups of topics, extensive experiments on the listed realtime PCR techniques are conducted during the course which also convey in a practical manner all essential knowledge of the individual techniques and areas of application, from planning through preparation and implementation to evaluation.
This course requires fundamental knowledge of PCR and basic molecular biological techniques.
